The inspiration for the song was "a cabin around Athens, Georgia," with a tin roof, where the band conceived "Rock Lobster", a single from their debut album. The cabin was said to have been located off the Atlanta Highway near Athens. It is based on a sample it uses from the Andrew Loog Oldham orchestral cover of the Rolling Stones' song "The Last Time", and involved some legal controversy surrounding a plagiarism charge. As a result, Mick Jagger and Keith Richards were added to the songwriting credits, and all royalties from the song went to former Rolling Stones manager Allen Klein.

Marc Bolan claimed to have written the song out of his desire to record Chuck Berry's "Little Queenie", and said that the riff was taken from the Berry tune. In fact, a slightly edited line (And meanwhile, I'm still thinking) from "Little Queenie" is said at the fade of "Get It On". According to producer Tony Visconti, this line was an unscripted ad-lib by Bolan during recording.

Some critics, such as Rolling Stone's Jon Landau, saw the song as a pastiche of John Lennon's sound, particularly the riff and the use of tape echo on the vocals. McCartney, however, didn't intend the song to be a pastiche of Lennon. He did say the vocal "does sound like John. ... I hadn't realised I'd sung it like John."

According to Randy Bachman, the inspiration for the song arose after an incident when he was visiting California. He was walking down the street with a stack of records under his arm, when he saw three "tough-looking biker guys" approaching. He felt threatened and was looking for a way to cross the street onto the other sidewalk when a little car pulled up to the men. A woman about 5 feet tall got out of the car, shouting at one of them, asking where he'd been all day, that he had left her alone with the kids, didn't take out the trash, and was down here watching the girls. The man was suddenly alone when his buddies walked away. Chastened, he got in the car as the woman told him before pulling away: "And one more thing, you ain't getting no sugar tonight". The words stuck in Bachman's memory.
